
Abstract
This project pioneers the establishment of prime editing in barley (Hordeum vulgare), a genome editing technique offering superior precision over conventional mutagenesis. Building on the CasCADE vector platform, the team will develop and optimize modular prime editing components—including guide RNAs and endonucleases—and validate their use in barley cells and whole plants. By editing agriculturally relevant traits such as stress tolerance, nutrient use, and plant architecture, the project aims to unlock precise and heritable trait improvements, contributing to resilient and sustainable barley breeding.
